The answer obviously depends on the person conducting the interview and the degree of complexity of the system you are being hired to build or maintain. You should inderstand subjects like the architectual structure of the system, distributing software, maintaining security and troubleshooting for example.
It will also help to look professional and have good communication skills both verbally and in writing. You may have an excellent understanding of the technology but, if you can't explain it clearly to the interviewer, you won't get the job.
